The Force Century Cycle Challenge, which starts and ends at Topsham Rugby Club, celebrates its 10th anniversary when the event is held on Sunday, June 28.
Money raised is donated to the Force cancer support centre at the Royal Devon and Exeter Hospital. The demanding route also involves a pedal across the tops of Exmoor.
Marc Millon, of Ferry Road, launched the biking challenge in 1999 in memory of his friend, Nello Ghezzo, who died of cancer that year.
Nello owned an Italian restaurant in Topsham.
Passionate cyclist Marc, 53, said: "Force does a fantastic job in researching cancer issues. The building of the support centre in Exeter was largely funded thanks to this event so we have close ties.
"The ride, known to many of us forever simply as 'The Nello' is one of the highlights of the annual cycling - and eating - calendar.
"It's an absolute classic, not just because the route is beautiful and challenging but because of the quality and quantity of delicious food on offer along the way."
